          研究方向:

            糖尿病是危害人類生命健康的一種重大代謝性疾病,我國是世界糖尿病第一大國,已成為我國公共健康面臨的最大挑戰之一。我們團隊圍繞胰島素分泌的分子機制開展了系統的研究,近期主要以機體中分泌胰島素的胰島器官為研究對象,通過建立和利用胰島類器官模型,研究人類胰腺的發育機制和糖尿病的發病機制。具體研究內容包括:

            1. 胰島素囊泡轉運和分泌的分子機制。以多個關鍵調控蛋白為切入點,利用生化和分子生物學、細胞生物學、蛋白質組學、基因編輯、活細胞超分辨成像、干細胞和類器官、動物模型等手段,研究胰島素囊泡在胰島beta細胞中生成和成熟、轉運和分泌、以及內吞和再循環過程的分子機制。

            2. 胰島類器官模型的建立和應用。利用干細胞、基因編輯、生物材料等多種技術,發展胰島類器官的誘導方法,獲得結構和功能更成熟的胰島類器官模型;并應用該模型,研究胰島的發育、組成和功能,以及糖尿病相關基因異常造成胰島發育缺陷和功能障礙的分子機制及其導致糖尿病的致病機制。
